    Latrunculin A delays anaphase onset in fission yeast by disrupting an ase1-independent pathway controlling mitotic spindle stability

    It has been proposed previously that latrunculin A, an inhibitor of actin polymerization, delays the onset of anaphase by causing spindle misorientation in fission yeast. However, we show that {Delta}mto1 cells, which are defective in nucleation of cytoplasmic microtubules, have profoundly misoriented spindles but are not delayed in the timing of sister chromatid separation, providing compelling evidence that fission yeast does not possess a spindle orientation checkpoint. Instead, we show that latrunculin A delays anaphase onset by disrupting interpolar microtubule stability. This effect is abolished in a latrunculin A-insensitive actin mutant and exacerbated in cells lacking Ase1, which cross-links antiparallel interpolar microtubules at the spindle midzone both before and after anaphase. These data indicate that both Ase1 and an intact actin cytoskeleton are required for preanaphase spindle stability. Finally, we show that loss of Ase1 activates a checkpoint that requires only the Mad3, Bub1, and Mph1, but not Mad1, Mad2, or Bub3 checkpoint proteins

    Most approaches to the formal analyses of cryptographic protocols make the perfect cryptography assumption, i.e. the hypothese that there is no way to obtain knowledge about the plaintext pertaining to a ciphertext without knowing the key. Ideally, one would prefer to rely on a weaker hypothesis on the computational cost of gaining information about the plaintext pertaining to a ciphertext without knowing the key. Such a view is permitted by the Generic Model and the Random Oracle Model which provide non-standard computational models in which one may reason about the computational cost of breaking a cryptographic scheme. Using the proof assistant Coq, we provide a machine-checked account of the Generic Model and the Random Oracle Mode

    An Application of The Solo Taxonomy to Classify Strategies Used by Pre-Service Teachers to Solve “One Question Problems”

    : The purpose of this article is to report on the strategies of secondary mathematics pre-service teachers (PSTs) as they solved conceptually rich problems. Using the Structure of Observed Learning Outcomes by Biggs and C (1982) (SOLO) Taxonomy, 15 PSTs’ solutions (in groups of 3 or 4) were analyzed by a panel of three mathematics educators. In addition, the authors studied questions posed by PSTs during their student teaching experiences through video analysis. Questions were then categorized using Crespo’s criteria of problem posing. Results showed a significant majority of the problems posed were procedural while PSTs own problem solutions showed a lack of conceptual understanding and depth of knowledge. The authors found the SOLO Taxonomy, together with PSTs scores on the state licensure exam and Crespo’s (2003) problem posing practices criteria to be a useful combination of tools to explore connections between PSTs’ mathematical and pedagogical content knowledge

    Enceladus: Cassini observations and implications for the search for life

    Aims. The recent Cassini discovery of water vapor plumes ejected from the south pole of the Saturnian satellite, Enceladus, presents a unique window of opportunity for the detection of extant life in our solar system. Methods. With its significant geothermal energy source propelling these plumes >80 km from the surface of the moon and the ensuing large temperature gradient with the surrounding environment, it is possible to have the weathering of rocks by liquid water at the rock/liquid interface. For the cases of the putatively detected salt-water oceans beneath the ice crusts of Europa and Callisto, an isolated subsurface ocean without photosynthesis or contact with an oxidizing atmosphere will approach chemical equilibrium and annihilate any ecosystems dependent on redox gradients unless there is a substantial alternative energy source. This thermodynamic tendency imposes severe constraints on any biota that is based on chemical energy. On Enceladus, the weathering of rocks by liquid water and any concomitant radioactive emissions are possible incipient conditions for life. If there is CO, CO2 and NH3 present in the spectra obtained from the plume, then this is possible evidence that amino acids could be formed at the rock/liquid interface of Enceladus. The combination of a hydrological cycle, chemical redox gradient and geochemical cycle give favorable conditions for life. Results. We discuss the search for signatures of these species and organics in the Cassini UVIS spectra of the plume and implications for the possible detection of life

    Effects of Extreme Obliquity Variations on the Habitability of Exoplanets

    We explore the impact of obliquity variations on planetary habitability in hypothetical systems with high mutual inclination. We show that large amplitude, high frequency obliquity oscillations on Earth-like exoplanets can suppress the ice-albedo feedback, increasing the outer edge of the habitable zone. We restrict our exploration to hypothetical systems consisting of a solar-mass star, an Earth-mass planet at 1 AU, and 1 or 2 larger planets. We verify that these systems are stable for 10810^8 years with N-body simulations, and calculate the obliquity variations induced by the orbital evolution of the Earth-mass planet and a torque from the host star. We run a simplified energy balance model on the terrestrial planet to assess surface temperature and ice coverage on the planet's surface, and we calculate differences in the outer edge of the habitable zone for planets with rapid obliquity variations. For each hypothetical system, we calculate the outer edge of habitability for two conditions: 1) the full evolution of the planetary spin and orbit, and 2) the eccentricity and obliquity fixed at their average values. We recover previous results that higher values of fixed obliquity and eccentricity expand the habitable zone, but also find that obliquity oscillations further expand habitable orbits in all cases. Terrestrial planets near the outer edge of the habitable zone may be more likely to support life in systems that induce rapid obliquity oscillations as opposed to fixed-spin planets. Such planets may be the easiest to directly characterize with space-borne telescopes.Comment: 46 pages, 12 Figures, 5 Table

    Most recent randomized controlled clinical trials have not found a benefit in antioxidants (vitamin C, vitamin E, and/or beta-carotene) for preventing cardiovascular disease, including stroke. These recent clinical studies have not confirmed earlier observational studies that suggested a benefit. No studies have assessed only stroke patients and stroke outcomes.
